choose scan program
wait for the machine come into “online” status
Setup: two things to be Setup
- cary option-mode/scan rate/wave length (simple read?)
- baseline: pick baseline correction, then ok
Go to the left bar-Calibration with the machine
set the zero (press zero button) to do Calibration.
blank sample means you calibrate with the solvent you will used in the procedures
when blank sample data comes out, press baseline button at the left bar
now the baseline of absorbance is set.
- After baseline set, message saying “block the sample beam and press ok to collect the 0 %T baseline scan”
- then block beam (use black plate to block the beam from the fright)
then the baseline is established from two aspects: absorbance and transmissivity (透射率)
- then remove the black card, load sample to measure. (the triangle side should be the left)
press “start” button to activate - remove the black card—-then add samples—–start
- After reading, click “ok”.
click “finish” to do save as—in your folder
- finished—-save methods (choose right format to save—CSV format)
tips
- Before and after, wash your cuvettes.
- When running, you can press the scale button/up and down to choose the right frame.
- when you wanna know the peak coordinates, press the peak label button to know.
- if your samples are concentrated, put sample first and then add solvent…
- add axis scale to zoom in what your sample is
